DropDownList.Items.Add method allows you to add new ListItem into the DropDownList. This item will be added as the last item of the DropDownList. DropDownList.Items.Insert method allows you to specify the index of the item within the DropDownList where you want to insert the ListItem.

DropDownList1.Items.Insert(0,"select"); here you have mandatory to give the index where you want to add. in this you can give the index position where you want to add the item. here i am adding 'select' in index position 0
